Sequence Analysis of a Mouse Secreted Superoxide Dismutase Gene in the Gracile Axonal Dystrophy (gad) Mouse


Abstract
The gracile axonal dystrophy(gad) mouse is an animal model for neurological disorders characterized by sensory ataxia followed by motor paresis of hind limbs. The mutation of gad mice is transmitted by a single autosomal recessive gene(gad)
located
on
proximal mouse chromosome 5. We recently found by fluorescent in situ hybridization on chromosomes and PCR analysis of mouse/hamster hybrid cells that a secreted superoxide dismutase(s-SOD) gene locates in a region close to gad locus. In the
present
study, we tested a hypothesis that altered expression of s-SOD causes gad phenotype using DNA sequencing and Northern blot analysis. cDNA encoding s-SOD was cloned and the nucleotide sequence was determined in the gracile axonal dystrophy(gad)
and
control mice. Both gad and control mice share identical s-SOD cDNA sequences. The sequence identity of deduced amino acid sequence between mouse s-SOD and rat epididymal SOD, and between mouse s-SOD and human extracellular SOD(SOD3) were 79 and
61%,
respectively. The size and the level of expression of gad s-SOD mRNA was not significantly different from that of the control. These findings suggest that the cause of gad mice is unlikely to be directly associated with the s-SOD gene.
